چکیده

Gartner, Theodore K. (University of California, Davis), and Monica Riley. Isolation of mutants affecting tryptophanase production in Escherichia coli. J. Bacteriol. 89:313-318. 1965.-Mutants of Escherichia coli K-12 were isolated which appear to have suffered an alteration in the regulation system governing tryptophanase synthesis. A novel selection method was used to isolate tryptophanase mutants from tryptophan synthetase deletion mutants. Mutants were obtained which exhibited the following phenotypes either singly or in combination with others: constitutivity and inducibility at 13 C (wild type is not inducible at 13 C), constitutivity and hyperinducibility at 37 C. Mutation to constitutivity at 13 C seems to require two mutational changes of the wild type. Other mutants, presumably structural gene mutations, were isolated which lack tryptophanase activity under all conditions tested.

Study of Mutations in the DNA gyrase gyrA Gene of Escherichia coli

Quinolones are a large and widely consumed class of synthetic drugs. Expanded-spectrum quinolones, like ciprofloxacin are highly effective against Gram-negative bacteria, especially Escherichia coli. In E. coli the major target for quinolones is DNA gyrase. This enzyme is composed of two subunits, GyrA and GyrB encoding by gyrA and gyrB, respectively. Mutations in either of these genes cause qu...

Unstable mutations that relieve catabolite repression of tryptophanase synthesis by Escherichia coli.

From strains of Escherichia coli that carry deletions of the trp region, five different mutants were isolated that were capable of synthesizing tryptophanase at unusually high rates in conditions of severe catabolite repression. Notwithstanding the comparative insensitivity to catabolite repression, the rates of tryptophanase synthesis in the mutants were greatly diminished by the introduction ...

Expression of tolC and organic solvent tolerance of Escherichia coli ciprofloxacin resistant mutants

AcrAB-TolC is a major efflux pump in Escherichia coli. It was reported that tolC is overexpressed and involves in improving the organic solvent tolerance level in Escherichia coli marR mutants that are resistant to several antibiotics, such as ciprofloxacin. Low and intermediate levels resistance did not improve organic solvent tolerance. Thus, in this descriptive-experimental study it was deci...
